Moderna, Inc. dropped $39.64 per share on Thursday, wiping out around $15.8 billion in market capitalization. Despite the 22.7% decline, its price of $134.74 stayed 63% higher than the analysts’ average target of $82.62. The U.S. markets remained open.

After a 177% surge on Wednesday driven by encouraging Phase 3 melanoma results, Moderna shares retreated. The stock had climbed to $174.38 from a projected $62.95 base. Thursday’s decline wiped out around 36% of the prior day’s increase, but about $28.7 billion of value remained.

The math is crucial. Investors are assigning a commercial value to a cancer platform ahead of any regulatory review of the drug. Analysts, for their part, have only just started to update their models in response to the trial outcome.

Moderna and Merck & Co., Inc. reported that intismeran autogene combined with Keytruda achieved both main goals in a Phase 3 trial. The study involved 1,137 participants and demonstrated gains in recurrence-free survival and distant-metastasis-free survival. Specific hazard ratio figures and overall survival results were not disclosed.

Karen Knudsen, CEO of the Parker Institute for Cancer Immunotherapy, described the outcome as “truly this next phase in immunotherapy.” Moderna President Stephen Hoge stated that the treatment could help eligible patients as early as next year, provided it is approved by regulators. Reuters

This program has the potential to be Moderna’s leading product outside of respiratory treatments. Barclays projects that yearly melanoma sales could approach $3 billion by 2035. Analysts at J.P. Morgan stated the rollout is key for the firm’s return to profitability.

The balance sheet offers management some breathing room. Moderna finished the second quarter expecting to end the year with between $4.7 billion and $5.2 billion in cash. The company also reduced its forecast for 2026 operating expenses by approximately $200 million.

The consensus remains guarded. According to Google Finance, there were two Buy ratings, 12 Hold, and one Sell. Of the latest targets listed, only Bank of America’s $170 price target was above Thursday’s closing price.

Trading levels remain exceptionally high. Approximately 85.7 million shares had traded by 15:30 EDT, representing 8.6 times Yahoo’s recent daily average. On Wednesday, volume was around 200 million shares, nearly half of the company’s total shares outstanding.

The high turnover suggests activity beyond basic buying. Short interest reached 13.7% before the shares surged. According to Brookline Capital Markets analyst Leah Cann, short covering added “a lot of paraffin on the flame.” The Wall Street Journal

Risks: Regulators require complete Phase 3 results. Scaling up personalized manufacturing poses challenges, pricing has yet to be determined, and the overall survival benefit is unclear. Delays in regulatory review or limited reimbursement could narrow the premium rapidly.

The next step for valuation is transparency, rather than more percentage figures. Investors are waiting for Phase 3 hazard ratios, safety information, and the expected timeline for regulatory filings. For now, the difference between analyst targets and current prices indicates the level of execution the market has already factored in.
