SION-719 was administered alongside Trikafta in a group of 15 adults with cystic fibrosis. Each participant carried two copies of the F508del mutation and was already on stable standard therapy.

The study found an average placebo-adjusted sweat chloride change of -1.0 mmol/L. The p-value stood at 0.7, indicating a lack of statistical significance.

Sionna pointed to atypical variability in sweat-chloride and inconsistent Trikafta exposure across periods, noting these issues are still being analyzed. However, these factors do not alter the unsuccessful endpoint outcome.

SION-719 was tolerated well over the 14-day period. The majority of adverse events were mild to moderate, and no serious adverse events were reported. RBC’s upgrade did not signal a bullish reversal, and its target dropped by 78% to $5. Remaining published targets now align closely with Sionna’s $5.93 gross cash per share number.

Risks are still high. SION-451 has data only from healthy volunteers. Sionna’s NBD1 approach is questioned by the failed patient trial. Gross cash may decline due to trials, contracts, leases, restructuring, and other obligations.

The upcoming key catalyst for the stock will be management’s announcement on its capital plan and the fate of SION-451 alongside SION-2222. Unless both are clearly valued, the difference between gross cash and current value serves as a buffer rather than supporting an investment case.
